Certain bikes have a reputation for being hard to work on. Take a quick look at the script beta.tcl. The blue residues are the LYS and GLY residues in the ubiquitin. You should see that the protein is mostly a collection of red spheres, with some residues shown in blue. ![]() In the Tk Console window, type sourcebeta.tcl and observe the color change. The script beta.tcl sets the colors of residues LYS and GLY to a different color from the rest of the protein by assigning them a different beta value, a trick you have already learned in Basic Protocol 6, steps 5–9. You should have downloaded a simple script, beta.tcl, with this unit. Just use any text editor to write your script file, and in a VMD session, use the command source filename to execute the file. When performing a task that requires many lines of commands, instead of typing each line in the Tk Console window, it is usually more convenient to write all the lines into a script file and load it into VMD. Thus, $crystal is now a function that performs actions on the contents of the “all” selection. This concept is particularly important when multiple molecules are loaded at the same time (see Basic Protocol 9 for dealing with multiple molecules in VMD). A top molecule means that it is the target for scripting commands. Instead of a molecule ID (which is a number), we have used the shortcut “top” to refer to the top molecule. ![]() This step creates a selection, crystal, that contains all the atoms in the molecule and assigns it to the variable crystal. The selection returned by atomselect is itself a command you will learn to use. The first argument to atomselect is the molecule ID (shown to the very left of the VMD Main window), the second argument is a textual atom selection like what you have been using to describe graphical representations in Basic Protocol 1. This command allows you to select a specific part of a molecule. Type set crystal in the Tk Console window. If you are using a Mac, your vmd console window is the terminal window that shows up when you open VMD.Ģ. The Tcl commands that you enter in the VMD TkConsole window can also be entered in the vmd console window. It should tell you a molecule has been loaded, as well as some of its basic properties like number of atoms, bonds, residues and etc. ![]() The vmd console window tells you what’s going on within the VMD session that you are working on. When you open VMD, by default a vmd console window appears. You can use the standard Unix commands in the VMD TkConsole window to navigate to the correct directory. If you see the error message ”Unable to load file ‘1ubq.pdb’ using file type ‘pdb’”, you might not be in the correct directory that contains the file 1ubq.pdb. In the VMD TkConsole window, type the command mol new 1ubq.pdb and hit enter.Īs you can see, this command performs the same function as described at the beginning of Basic Protocol 1, namely, loading a new molecule with file name 1ubq.pdb. ![]() ![]() Renesas is a global leader in microcontrollers, analog, power and SoC products and provides developers with a significant boost in processing power, memory, and functionality.īased on an Arm® Cortex®-M4 core, the RA4M1 microcontroller on the Arduino UNO R4 features a clock speed of 48 MHz for higher processing power. Each of these analog pins has an inbuilt ADC of resolution of 2 10 bits (so it will give 1024 values). The Leonardo consists of 12 analog inputs, labeled from A0 to A11, all of these pins can also be used as digital I/O pins. The story was way too elbaorte and far too much was revealed about the orchestrators of this torture world. Ultimately, the movie went too big out of the gate. This is really a more PG version of Saw with no moral compass. The idea is interesting and there's some clever twists and turns. It works enough that I wouldn't mind more, if the filmmakers get their priorities right.Įscape Room feels like watching part 3 or 4 of a franchise where everyone starts noting the formula and idea has lost its edge. Much like The Maze Runner, once the characters are outside the maze, I just don't care. It's a relatively minor but enjoyable thriller that lives in the moment and, in the future, should find more time to spend in the rooms and the crazy challenges. Once it extends beyond the rooms and tries building a mythology and a conspiracy behind the rooms, that's where Escape Room starts to fall apart. The movie is fun as you're watching it as long as you accept that little will make sense. ![]() The assorted characters have a personal connection that is flimsy and leads to a late reveal of who is responsible for this wicked game. The fun doesn't come from the puzzles but more what the rooms will do next. They mostly involve clues that won't make sense because we can't explore the room, so it's just waiting for characters to solve things and make connections. The puzzles aren't exactly things that an audience can solve in the moment. The production design is inventive and impressive for its minor budget. The enjoyment of the movie is the curiosity of the rooms and the various challenges. “Thinning out excess plants is important to develop good-sized onion bulbs, but often I will thin out crowded onion plants gradually and harvest the larger than normal thinnings to eat like a spring onion,” shares Master Horticulturist Dan Ori. ![]() With outdoor sown onions, these can now be further thinned to a final spacing as above, depending on the desired size of the harvestable crop. However for smaller onions, to maximise space, this distance can be reduced. Plant out the onions at the same depth they were previously planted and water in.įor decent-sized onions it is recommended to plant them 10–15 cm apart. On this occasion, our seedlings were grown undercover in trays in a greenhouse, with water added regularly to the bottom of the tray. Having been grown on in small pots undercover, the onions can be planted out, but only having been gradually hardened off first. With indoor sown onions, thin the seedlings to 1–3 per module and when they are 5-7cm tall, prick out into individual pots to grow on under cover.įor outdoor sowings, once the seedlings are large enough to handle, they can be thinned to a spacing of 5–7cm, or less for more onions of a smaller size. If sowing directly outdoors, prepare the soil by raking to a fine tilth and sow the seed thinly in rows 15-20cm apart at a depth of 1cm.Ĭarefully water after sowing and keep the soil moist, but not wet. Water gently on sowing to avoid dispersing the seed and keep at a minimum temperature of 10☌, with the soil moist until germination occurs. ![]() If sowing onion seed indoors, fill a seed or module tray with a seed sowing compost mix and sow the seeds individually in cells or in groups of 3–4 at a depth of 1cm and cover over with compost. Onion seed is cheaply and readily available and is therefore most often purchased rather than being collected. Store the seed in a cool, dry place to sow the following spring. To harvest onion seed, allow a stem to develop flowers and subsequently set seed before allowing to dry and collecting using a paper bag. Onions can also be sown outdoors later in spring, but only once the soil has begun to warm up. The months of January and February are generally considered the best. ( Update: The PP export had all my passwords, but only ~380 were imported into LastPass. This means I have to manually double check every one of my ~500 records to make sure everything has been captured. They likely got lost in the conversion from PP to 1Password, and so don’t appear in LastPass either. ![]() Some records just didn’t import at all.Such ‘tags’ are searchable as part of the full-text search capability in LastPass, but as they are entered manually, there’s no master list of tags to choose from as you start to enter one (1Password has tagging capability). Each record was tagged with the category it had in PP, but I can’t see any way to add tags to LastPass, except manually in the Notes field. NOTE: Almost all the imported records went in as Notes records (called Secure Notes in 1Password), with limited splitting out into fields. However, in my testing for 1Password, I’d converted a PP CSV file to 1Password’s *.1PIF file and that imported fine into LastPass.PP was NOT an import option type for LastPass, and the CSV file I’d exported from PP had NO records that LastPass recognised when I tried to import it.For those that I could add fields to, when you add them, you can’t rearrange them in the order you want, so you have to get them right first. might have this ability, but I need to test further. Because I already had this account, I wasn’t asked to enter payment credentials, though I’m not sure that’s the case for totally new users.) My first tests were to see how easy the interface was to use, whether I could import my Passwords Plus (PP) data, what sort of template/categories it used, whether I could customise fields/field labels to suit my data etc. I didn’t download the Android app for it, but I did install the browser extension (it was required, so I couldn’t avoid that). That said, my next test, and the topic of this post was LastPass, which is a web-based app.Īs for 1Password, I only downloaded/installed the free trial of LastPass and tested some of the functionality that I wanted. I haven’t ruled out 1Password yet as it does have a Windows app, and that’s what I’m used to.
